About Amifostine
Antidote
Uses
To reduce the neutropenia related risk of infection due to cyclophosphamide and cisplatin in patients with advanced ovarian carcinoma. Protect against cumulative nephrotoxicity of cisplatin containing regimens in patients with advanced solid tumours of non-germ cell origin. Protect against acute and late xerostomia in head and neck cancer in association with radiation therapy.
How to take it
Adults: 910 mg/m2 once daily as a 15 min IV infusion starting within 30 min prior to chemotherapy given by short infusion. If full dose cannot be given reduce to 740 mg/m2 for subsequent cycles; see literature. To protect against xerostomia, 200 mg/m2 once daily as a 3 min IV infusion starting within 15-30 min prior to radiotherapy. Children: Not applicable. For IV infusion. Keep patients in a supine position during the infusion. When used prior to chemotherapy, monitor BP every 5 min during the infusion and thereafter as clinically indicated.
