About Pyrazinamide
Antituberculosis Preps
Uses
Treatment of tuberculosis only in conjunction with other anti-tuberculous drugs.
How to take it
Adults & Children: 15-30 mg/kg daily in divided doses; max 2 gm daily.
Side effects
Hepatitis which can be minimized by dosage adjustment. Hepatic reactions, active gout, sideroblastic anaemia, arthralgia, hyperuricaemia, anorexia, GI upset, flushing, dysuria, malaise, fever, rash, aggravation of peptic ulcer, wt loss, photosensitivity.
Pregnancy and breastfeeding
Pregnancy: Risk cannot be ruled out. Lactation: Caution advised or effect undetermined. Contraindications: Liver disease, hyperuricaemia or gouty arthritis, acute porphyria. Precautions: Perform frequently liver function tests and blood uric acid determinations. History of gout or diabetes. Withdraw treatment if liver damage or hyperuricaemia accompanied by acute gouty arthritis occurs. Renal impairment.
Advice
Take with meals. Contact prescriber if you experience colour change in urine, pale stools, difficulty in urination, blood in urine, easy bleeding or bruising, yellowing of skin or eyes, extreme joint pain, severe pain in feet or toes, unrelieved nausea or vomiting, unusual fever. Regularly follow-up medical and laboratory tests as advised by prescriber. Avoid direct sunlight, wear sunscreen, proper eyewear and protective clothing to prevent photosensitivity.

Interactions with Pyrazinamide
Probenecid, sulfinpyrazone, oral typhoid vaccine, combined hormonal contraceptives, alcohol, aspirin, ascorbic acid, hypoglycaemics, rifampicin, iodinated contrast agents.
