Indications

Treatment of noncirrhotic nonalcoholic steatohepatitis (NASH) with moderate to advanced liver fibrosis (consistent with stages F2 to F3 fibrosis).

Dosage

Adults: Body wt <100 kg: 80 mg once daily. =100 kg: 100 mg once daily. Children: Not recommended.

Adverse Effects

Diarrhoea, nausea, vomiting, dizziness, pruritus, abdominal pain, constipation.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Risk cannot be ruled out. Lactation: Caution advised or effect undetermined. Contraindications: Decompensated cirrhosis. Moderate to severe hepatic impairment (Child-Pugh Class B or C). Precautions: Monitor patients during treatment for elevation in liver tests and for the development of liver-related adverse reactions. Acute cholecystitis, cholelithiasis. Elderly.

Interactions

Strong or moderate CYP2C8 inhibitors (e.g. gemfibrozil, clopidogrel etc); avoid. OATP1B1 and OATP1B3 inhibitors (e.g. cyclosporine); avoid. CYP2C8 substrates. Atorvastatin, Pravastatin, Rosuvastatin and Simvastatin.

Advice to Patient

Take with or without meals. Immediately report any signs or symptoms of severe liver injury (e.g. fatigue, nausea, vomiting, right upper quadrant pain or tenderness, jaundice, fever, rash) or symptoms of gallbladder adverse reactions, including cholelithiasis, cholecystitis, and obstructive pancreatitis (gallstone).
