Indications
Advanced renal cell carcinoma (RCC) after failure of one prior systemic therapy.
Dosage
The starting dose is 5 mg orally twice daily. Dose adjustments can be made based on individual safety and tolerability.
Adverse Effects
The most common (=20%) adverse reactions are diarrhoea, hypertension, fatigue, decreased appetite, nausea, dysphonia, palmar-plantar erythrodysesthesia (hand-foot) syndrome, weight decreased, vomiting, asthaenia and constipation.
Risk Factor (Pregnancy/Lactation)
Precautions: Hypertension, arterial and venous thrombotic, Hemorrhagic events.
Interactions
Dexamethasone, bosentan, modafinil, St. John's Wort (Hypericum perforatum), or medicine for asthma, tuberculosis, seizures, bacterial infections, fungal infections, depression, or HIV/AIDS.
Advice to Patient
Take with or without meals. Swallow tablet whole with water. Contact prescriber if there is severe diarrhoea, nosebleed, bleeding from gums, headache, muscle ache, cough, sore throat, difficulty in urination, cold, shivering. Take plenty of fluids during therapy. Avoid grapefruit and grapefruit juice during therapy. Avoid immunisation with live vaccines during treatment and for 6 months after completion of treatment.

Absorption: Rapidly absorbed from the GI tract. Bioavailability: Approx. 58%. Metabolism: Hepatic. Half-life elimination: 2.5-6.1 hrs. Time to peak, plasma: 2.5-4 hrs. Excretion: Faeces (approx. 41%). Urine (approx. 23%).
