About Dapoxetine

Antidepressant

Uses

Premature ejaculation (ejaculation with minimal sexual stimulation and an intravaginal ejaculatory latency time <2 mins) in the majority of intercourse attempts over the preceding 6 months and causing marked personal distress or interpersonal difficulty.

How to take it

Adults: Max one dose in 24 hrs. Initially 30 mg as needed 1-3 hrs before sexual activity. If response inadequate and no adverse reactions or no symptoms of syncope occur, the dosage may be increased to 60 mg if needed. Evaluate benefit after 4 wk or 6 doses, re-evaluate every 6 mth. Children: Not recommended.

Side effects

Dizziness, headache. GI upset, anxiety, agitation, fatigue, irritability, sleep disorders, decreased libido, somnolence, attention disturbance, tremor, paraesthesia, blurred vision, tinnitus, flushing, sinus congestion, yawning, dry mouth, hyperhidrosis, erectile dysfunction, increased BP. Withdrawal symptoms.

Pregnancy and breastfeeding

Contraindications: Significant cardiac conditions, psychiatric disorders including depression and history of mania/hypomania, unstable epilepsy. Elderly. Renal or hepatic impairment. Precautions: Advise patients to report distressing feelings. Risk of orthostatic hypotension and syncope, warn patient and advise adequate hydration. Stable epilepsy. Raised intra-ocular pressure, patients at risk of closed angle glaucoma. Bleeding disorders. CYP2D6 poor metabolisers. Mild to moderate renal impairment.

Advice

Take with or without meals. Swallow whole with a full glass of water. Avoid alcohol during therapy. Do not take this medication if you are dehydrated. Contact prescriber if you experience seizures, light-headedness, changes in mood, suicidal thoughts.

Interactions with Dapoxetine

Other CNS drug, St. John's Wort and other serotonergic agents. Vasodilators, anticoagulants, drugs affecting platelet function, PDE5 inhibitors, CYP3A4 inhibitors (e.g. ketoconazole, ritonavir), potent CYP2D6 inhibitors, alcohol, MAOIs, SSRIs, SNRIs, TCADs, alcohol.
