Uses

Treatment of advanced or metastatic renal cell carcinoma. Treatment of unresectable or metastatic gastro-intestinal stromal tumours (GIST). Treatment of unresectable or metastatic pancreatic neuroendocrine tumours.

How to take it

Adults: Treatment of advanced or metastatic renal cell carcinoma. Treatment of unresectable or metastatic gastro-intestinal stromal tumours (GIST): 50 mg daily for 4 wk, followed by a 2 wk treatment-free period to complete 6 wk cycle: adjust dose in steps of 12.5 mg according to tolerability: dose range 25-75 mg/day. Treatment of unresectable or metastatic pancreatic neuroendocrine tumours: 37.5 mg once daily without treatment-free period; adjusted in steps of 12.5 mg, doses adjusted according to tolerability. Max: 50 mg/day. Children: Not recommended.

Pulmonary embolism, haemorrhage, neutropenia, anaemia, thrombocytopenia, leucopenia, lymphopaenia, headache, hypertension, GI upsets, rash, insomnia, skin disorders, alopecia, infections, blood dyscrasias, hypothyroidism, anorexia, dehydration, hypoglycaemia, insomnia, depression, dizziness, dysgeusia, peripheral neuropathy, paraesthesia, hypoaesthesia, hyperaesthesia, lacrimation, oedema, thromboembolism, flushing, hot flush, dyspnoea, cough, pleural effusion, oropharyngeal pain, nasal congestion or dryness, dry mouth, dysphagia, cheilitis, proctalgia, skin, hair and nail discolouration (warn patient), skin and nail disorders, pain (incl chest, musculoskeletal, extremity), muscle spasm and…

Pregnancy and breastfeeding

Pregnancy: Risk cannot be ruled out. Lactation: Caution advised or effect undetermined. Contraindications: Severe hepatic impairment. Precautions: Risk of fatal pulmonary haemorrhage, hypertension. Perform complete blood count prior to treatment. Pre-existing cardiac disease, electrolytes disturbances, monitor for thyroid dysfunction, pancreatitis and heart failure. History of, or risk factors for cardiac events or QT prolongation. Diabetes. Perform baseline urinalysis and monitor for development or worsening of proteinuria; discontinue if nephrotic syndrome occurs. Monitor LFTs. Interactions with Sunitinib

Rifampicin, phenobarbital, phenytoin, carbamazepine, dexamethasone, anticoagulants, St John's wort or other CYP3A4 inducers, antifungals, ritonavir, macrolides, grapefruit juice or other CYP3A4 inhibitors, antiarrhythmics or other drugs that prolong QT interval, IV bisphosphonates.
