Antipeptic Ulcerant

Uses

Benign gastric, duodenal or NSAID-associated ulceration, gastro-oesophageal reflux disease.

How to take it

ORAL: Adults: Benign gastric, duodenal or NSAID-associated ulceration: 300 mg in the evening or 150 mg twice daily for 4-8 wk, maintenance, 150 mg at night. Gastro-oesophageal reflux disease: 150-300 mg twice daily for 12 wk. Children: =12 yrs: Oral Soln: 150 mg twice daily for up to 8 wk. Max 300 mg daily. INJECTION: Adults: By IV, for short term use in peptic ulcer, by intermittent IV infusion over 15 min, 100 mg three times daily, by continuous IV infusion, 10 mg per hr; max 480 mg daily. Children: Not recommended. For intermittent IV infusion, administer over 15 min. Compatibility: Glucose 5% or NaCl 0.9%.

Side effects

Diarrhoea, dizziness, rash, tiredness. Gynaecomastia, occasional reversible liver damage, confusion. Very rarely interstitial nephritis, acute pancreatitis, thrombocytopenia, leucopenia, pancytopenia, aplastic anaemia, fever, headache, myalgia, arthralgia, sinus bradycardia, tachycardia, heart block, anaphylaxis, hallucinations, depression, sweating, urticaria, raised liver enzyme, hepatitis, jaundice, somnolence, rarely hypersensitivity reaction.

Pregnancy and breastfeeding

Pregnancy: Risk cannot be ruled out. Lactation: Contraindicated or not recommended. Precautions: Exclude malignant disease before and during treatment. Impaired renal function. Monitor patients on long term therapy.

Advice

Take 1 hour before meals. Do not take within 1 hour of an antacid. Report prescriber immediately about abdominal pain, muscle or joint pain, easy bruising, extreme fatigue, yellow skin or sclera, difficulty in swallowing food, bloody vomit, or bloody or delayed stools, fever, sore throat. Drink plenty of fluids (if allowed), eat high-fiber foods and exercise regularly to minimize constipation. Avoid smoking during therapy.
