Uses

Erectile dysfunction. Erectile dysfunction in patients on alpha-blocker therapy.

How to take it

Erectile dysfunction: Adults: Initially 100 mg, to be taken approx. 15-30 mins before sexual activity, then altered acc to response to 50-200 mg (max per dose 200 mg), to be taken as a single dose as required. Max: 1 dose daily. Erectile dysfunction in patients on alpha-blocker therapy: Adults: Initially 50 mg, to be taken approx. 15-30 mins before sexual activity, then altered acc to response to 50-200 mg (max. per dose 200 mg), to be taken as a single dose as required. Max: 1 dose daily. With concurrent use of moderate CYP3A4 inhibitors: Max 100 mg once every 48 hrs.

Side effects

Headache, flushing, nasal congestion.

Pregnancy and breastfeeding

Contraindications: Females. Severe renal (CrCl <30 mL/min) or hepatic impairment. Cardiac conditions where sexual activity is inadvisable, MI, stroke or life-threatening arrhythmia in the previous 6 months, resting BP <90/50 mmHg or >170/100 mmHg, unstable angina, CHF (NYHA class II-IV). Hereditary degenerative retinal disorders, non-arteritic anterior ischaemic optic neuropathy. Precautions: Cardiovascular disease, left ventricular outflow obstruction, mild to moderate hepatic impairment. Abnormal penile anatomy or predisposition to priapism (e.g., sickle cell anaemia, multiple myeloma or leukaemia). Bleeding disorders, active peptic ulcer.

Advice

Take with or without meals. Take approx. 15-30 mins before sexual activity. Report immediately any changes in vision or hearing, or erection lasting >4 hrs.

Interactions with Avanafil

Nitrates, amyl nitrite, other erectile dysfunction agents, alcohol, antihypertensives, CYP3A4 inhibitors (e.g., aprepitant, macrolides, HIV protease inhibitors, azole antifungals, nefazodone), a-blockers, grapefruit juice, CYP inducers, riociguat.
