About Omeprazole

Antipeptic Ulcerant

Uses

Oesophageal reflux disease, reflux oesophagitis. Long-term management of acid-reflux disease. Duodenal and benign gastric ulcers including those complicating NSAID therapy. Duodenal ulcers associated with H. pylori. Zollinger-Ellison syndrome. Prophylaxis of acid aspiration during general anaesthesia.

How to take it

Adults: ORAL: Duodenal ulcer: 20 mg once daily for 4 wk. Severe cases, increase to 40 mg once daily. Prevention of relapse, 20 mg once daily, dose may range between 10 mg to 40 mg daily. Gastric ulcer: 20 mg once daily for 8 wk. Severe cases, increase to 40 mg once daily. Prevention of relapse, 20 mg once daily, increased to 40 mg once daily if necessary. Acid-reflux disease: Long-term management, 10 mg once daily, increasing to 20 mg once daily if symptoms return. GERD: 20 mg once daily for 4 wk, extend for a further 4-8 wk if not fully healed. Side effects

Headache, GI upset, skin rashes, paraesthesia, dizziness, somnolence, insomnia, arthralgia, myalgia. Increased risk of GI infections. Increase in liver enzymes, thrombocytopenia, fundic gland polyps. Report of subacute cutaneous lupus erythematosus.

Pregnancy and breastfeeding

Pregnancy: Risk cannot be ruled out. Lactation: Contraindicated or not recommended. Precautions: Exclude malignancy before treatment. Hepatic impairment. Monitor serum B12 in severely ill children on long term treatment. Possible increased risk of GI infection.

Advice

Take before meals, best if taken before breakfast. Swallow whole. Do not crush or chew capsules. If there is difficulty in swallowing, open the capsule and mix capsule contents in 1 tablespoon applesauce. Do not chew, swallow immediately with a glass of water. For oral suspension, mix the packet contents in 2 tablespoon of water in a small cup, stir it well, drink immediately. Drink a cup of water again. Contact prescriber if there is abdominal pain, diarrhoea, testicular pain, changes in urination, pain on urination or changes in respiratory status. During treatment avoid alcohol, aspirin products, ibuprofen and foods that may increase gastric secretions.

Interactions with Omeprazole

Diazepam, phenytoin, warfarin, digoxin, ketoconazole, itraconazole, atazanavir, clopidogrel, erlotinib, cilostazol, saquinavir, tacrolimus, methotrexate, inducers or inhibitors of CYP2C19 or CYP3A4.
