Tegaserod
Laxative · GI serotonin receptor agonist (GI-SRA)
Indications
Treatment of constipation predominant irritable bowel syndrome in patients whose main symptoms are constipation, abdominal pain and discomfort.
Dosage
Adults: 6 mg twice daily prior to a meal for 12 wk. Children: Not recommended.
Adverse Effects
Gl complaints (e.g. diarrhoea, abdominal pain, nausea, flatulence), headaches, dizziness, back pain and influenza-like symptoms.
Risk Factor (Pregnancy/Lactation)
Pregnancy: No evidence of risk in humans. Lactation: Contraindicated or not recommended. Contraindications: Hypersensitivity to tegaserod. Precautions: Special care is necessary in patients with severe hepatic impairment and in patients with lactose intolerance.
Advice to Patient
Take on an empty stomach at least 30 minutes before meals. Contact prescriber if severe diarrhoea, abdominal cramps or increased abdominal pain occurs.

Absorption: Rapidly absorbed from the GI tract. Metabolism: Hydrolysis and direct glucuronidation. Bioavailability: Oral: Absolute: 10%. Half-life elimination: 11 hrs. Time to peak, plasma: After 1 hr. Excretion: Faeces (2/3rd of the dose as unchanged drug). Urine (1/3rd of the dose as metabolites).
