Indications

Moderate to severe atopic dermatitis unresponsive or intolerant to other therapies.

Dosage

Adults: Apply thin layer of 0.1% oint twice daily until lesions clear. Children: Under 2 yr, not recommended; 2-16 yr, apply thin layer of 0.03% oint twice daily for upto 3 wk, then reduce to once daily and continue until condition cleared.

Adverse Effects

Burning, pruritus, erythema, sensitivity, tingling, alcohol intolerance, increased risk of folliculitis, acne and herpes simplex.

Risk Factor (Pregnancy/Lactation)

Contraindications: Hypersensitivity to macrolides, genetic epidermal barrier defects. Pregnancy, lactation. Precautions: Hepatic failure, generalized erythroderma. Minimize exposure to UV light. Treat infections before therapy. Monitor patients who develop lymphadenopathy. Avoid eyes and mucous membranes.

Interactions

Avoid use of emollients within 2 hours. CYP3A4 inhibitors in widespread or erythrodermic disease, vaccines.
