Indications

Prevention and treatment of ENT. Bronchopulmonary, cutaneous, mouth and gonococcal genital infections. Toxoplasmosis in pregnancy.

Dosage

Adults: 500 mg every 8 hr or every 12 hr. Children: > 20 kg: 500 mg-1 gm/10 kg body wt/day in 2-3 divided doses. Prophylaxis of meningococcal meningitis: Adults: 1000 mg every 12 hr for 5 days. Children: > 20 kg: 250 mg/10 kg body wt every 12 hr for 5 days.

Adverse Effects

Gl disturbances. Allergic reactions. Transient paraesthesia, pseudomembranous colitis, angioedema, liver function test abnormalities.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Risk cannot be ruled out. Lactation: Contraindicated or not recommended.

Advice to Patient

Take with or without meals.

Pharmacokinetics

Absorption: Incompletely absorbed from the GI tract. Absorption is decreased by the presence of food. Metabolism: Hepatic. Bioavailability: Absolute: 10%-60%. Half-life elimination: 5-8 hrs. Time to peak, plasma: 1.5-3 hrs. Excretion: Urine (10%). Faeces (mainly).
