Indications

Spinal muscular atrophy in paediatric and adult patients.

Dosage

ORAL: To be taken once daily after a meal via the provided oral syringe. <2 mth: 0.15 mg/kg. 2 mth-<2 yrs: 0.2 mg/kg. =2 yrs and wt <20 kg: 0.25 mg/kg. =2 yrs and wt =20 kg: 5 mg.

Administration

Caution should be exercised in the handling of Risdiplam powder for oral soln. Avoid inhalation and direct contact with skin or mucous membranes with the dry powder and the constituted soln. If such contact occurs, wash thoroughly with soap and water, rinse eyes with water. Wear disposable gloves during the preparation and cleanup procedure. Preparation of soln: 1. Gently tap the bottom of the closed glass bottle to loosen the powder. 2. Remove the cap. Do not throw away the cap. 3. Carefully pour 79 mL of Purified Water into the Risdiplam bottle to yield the 0.75 mg/mL oral soln. 4. Insert the Press-In bottle adapter into the bottle opening by pushing it down against the bottle lip. Ensure it is completely pressed against the bottle lip. 5. Adverse Effects

Fever, diarrhoea, rash, upper RTI, lower RTI, constipation, vomiting and cough.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Contraindicated in pregnancy. Lactation: Caution advised or effect undetermined.

Interactions

Drugs that are substrates of multidrug and toxin extrusion (MATE) transporters (Avoid).

Advice to Patient

Dose must be taken immediately after it is drawn up into the oral syringe. If not taken within 5 mins, discard drug from the oral syringe, and a new dose should be prepared. In breastfed infants, drug should be given after breastfeeding session. It cannot be mixed with formula or milk. Drink water after taking drug to ensure complete swallowing. If a dose is missed, administer within 6 hrs of the missed dose, resume the usual dose on the next day.
