Indications

Chronic indications: Brain ageing, psycho-organic syndrome, cerebral atherosclerosis, cerebral circulatory insufficiencies, brain ischaemia. Sub-acute indications: Psychic, motor and sensory rehabilitation ion (hemiplegia, aphasia). Acute indications: Comas, cerebral vascular accidents, head injuries, brain intoxications, intensive care and resuscitation.

Dosage

ORAL: Adults: Initially 7.2 gm daily, increase, if necessary, in increments of 4.8 gm daily at three or four day intervals; max 20 gm daily in 2-3 divided doses. Children: Not recommended. INJECTION: Adults: 1 gm IV 3 times per day. Children: 30-50 mg/kg/day. Sub-acute indications: Adults: 3 amp twice daily. Acute indications: Adults: 3 amp four times daily.

Administration

Administered IV at the recommended daily dose. The injectable ampoules administered IV over several mins. The infusion is administered continuously at the recommended daily dose over a 24 hr period.

Adverse Effects

Hyperkinesia, insomnia, weight gain, somnolence, nervousness. Depression, diarrhoea, rash.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Contraindicated in pregnancy. Lactation: Contraindicated or not recommended. Contraindications: Severe renal insufficiency, hepatic impairment, cerebral haemorrhage. Precautions: Renal impairment. Elderly. Withdraw therapy gradually. Monitor patients for suicidal thoughts or behaviour.

Interactions

Thyroid hormones, St John's wort.

Advice to Patient

Take before meals.

Absorption: Rapidly and extensively absorbed from the GI tract. Bioavailability: Oral: Approx. 100%. Half-life elimination: 5 hrs. Time to peak, plasma: Oral: 1.5 hrs. Excretion: Urine.
