Moclobemide

Antidepressant · Reversible MAO-A inhibitor

Indications

Major depression.

Dosage

Adults: 300 mg daily in divided doses after meals. Severe depression 600 mg/day. Children: Not recommended.

Adverse Effects

Transient sleep disturbance, dizziness, nausea, headache, confusional states, restlessness, agitation.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Positive evidence of risk: Use only when no safer alternative exists for a serious problem. Lactation: Caution advised or effect undetermined. Contraindications: Acute confusional states, pheochromocytoma. Precautions: Severe hepatic impairment, thyrotoxicosis. Patients with excitation or agitation as the predominant clinical feature or schizophrenia. Avoid excessive amounts of tyramine rich food.

Interactions

5-HT reuptake inhibitors (including those which are tricyclic antidepressants), indirectly acting sympathomimetic amines, cimetidine, pethidine, codeine, morphine, fentanyl.

Advice to Patient

Take with meals or just after a meal. If diarrhoea occurs, drink plenty of water. Contact prescriber if there is difficulty in sleeping, restlessness or irritability, tingling, rash or swelling.

Absorption: Readily absorbed from the GI tract. Metabolism: Hepatic (first-pass). Bioavailability: Absolute: Approx. 55%-90%. Half-life elimination: 2-4 hrs. Cirrhosis: 4 hrs. Time to peak, plasma: 1 hr. Excretion: Urine.
