Hypolipidaemic/Anti-atheroma · small interfering RNA (siRNA) molecules
Indications
As an adjunct to diet and maximally tolerated statin therapy for the treatment of adults with heterozygous familial hypercholesterolaemia (HeFH) or clinical atherosclerotic cardiovascular disease (ASCVD), who require additional lowering of low-density lipoprotein cholesterol (LDL-C).
Dosage
Adults: Initially, 284 mg administered as a single SC inj, again at 3 months, and then every 6 months, to be given in combination with maximally tolerated statin therapy. Children: Not recommended.
Administration
To be administered via SC route into the abdomen, upper arm, or thigh. To be administered by a healthcare professional. Stability: Store at controlled room temperature 20-25°C with allowable excursions between 15-30°C.
Adverse Effects
Inj site reactions, arthralgia, urinary tract infections, diarrhoea, bronchitis, pain in extremity, and dyspnoea.
Risk Factor (Pregnancy/Lactation)
Pregnancy: Contraindicated in pregnancy. Lactation: Contraindicated or not recommended. Precautions: Severe renal or hepatic impairment.
Advice to Patient
Female patients of reproductive potential must use effective contraception during treatment.
Pharmacokinetics
Metabolism: Primarily metabolized by nucleases to shorter nucleotides of varying length. Half-life elimination: Terminal: Approx. 9 hrs. Time to peak, plasma: 4 hrs.
