Indications
Used as long-term enzyme replacement therapy in patients with Type 1 Gaucher disease (resulting in either anaemia, thrombocytopaenia, bone disease, hepatomegaly or splenomegaly).
Dosage
Adults and Children: Administered via IV infusion over 1-2 hrs. Dosage should be individualized to each patient. Initial dosages range: 2.5 U/kg of body wt. 3 times a wk - 60 U/kg once every 2 wks. Dosage must be adjusted on an individual basis. Children <2 yrs: Not recommended.
Administration
Administered via IV infusion over 1-2 hrs. Aseptic techniques should be used when diluting the dose. Reconstitution: A nominal 5 mL for the 200 unit vial (10 mL for the 400 unit vial) is withdrawn from each vial. The appropriate amount of Cerezyme for each patient is diluted with 0.9% NaCl inj USP, to a final vol of 100-200 mL. After reconstitution, Cerezyme should be inspected visually before use. The diluted soln may be filtered through an in-line low protein binding 0.2 µm filter during administration. Stability: Any vials exhibiting opaque particles or discolouration should not be used. Cerezyme does not contain any preservative, after reconstitution, vials should be promptly diluted and not stored for subsequent use. After reconstitution, at room temp (25°C) and 2-8°C it is stable for up to 12 hrs. When diluted, it is stable for up to 24 hrs at 2-8°C.
Adverse Effects
Nausea, abdominal pain, vomiting, diarrhoea, rash, fatigue, headache, fever, dizziness, chills, backache, tachycardia.
Risk Factor (Pregnancy/Lactation)
Precautions: Previous treatment with alglucerase injection. Pregnancy, lactation.

Half-life elimination: 3.6-10.4 mins. Time to peak effect: 30 mins.
