Cytostatic · Oestrogen antagonist
Indications
Treatment of oestrogen-receptor-positive metastatic or locally advanced breast cancer in postmenopausal women in whom disease progresses or relapses while on, or after, other anti-oestrogen therapy.
Dosage
Adults: By deep intramuscular injection into buttock, 500 mg every 2 weeks for the first 3 doses and then 500 mg every month. 500 mg dose should be administered as one 250 mg injection (slowly over 1-2 minutes) into each buttock. Children: Not recommended.
Administration
Administered by slow IM injection (1 to 2 minutes per injection) into the buttocks as two 5 mL injections, one in each buttock. Stability: Refrigerate, 2°C to 8°C (36°F to 46°F. Store in the original package to protect from light.
Adverse Effects
Nausea, vomiting, diarrhoea, venous thromboembolism, anorexia, rash, hypersensitivity reactions, hot flushes, back pain, less commonly vaginal hemorrhage, headache, asthaenia, urinary tract infections, vaginal candidiasis and leucorrhoea.
Risk Factor (Pregnancy/Lactation)
Pregnancy: Contraindicated in pregnancy. Lactation: Contraindicated or not recommended. Contraindications: Severe renal or hepatic impairment. Precautions: Mild to moderate renal or hepatic impairment. CrCl < 30 mL/min as no information is available.
Pharmacokinetics
Metabolism: Via combinations of a number of possible biotransformation pathways analogous to those of endogenous steroids. Excretion: Faeces (approx 90%). Urine (negligible; < 1%).
