Flupenthixol and Melitracen

Antidepressant · Thioxanthine/Tricyclic antidepressant (TCAD)

Indications

Anxiety, depression, apathy.

Dosage

Adults: Usually 2 tab daily morning and noon. In severe cases the morning dose may be increased to 2 tab. Elderly: 1 tab in the morning. Maintenance dose: Usually 1 tab in the morning. Children: Not recommended.

Adverse Effects

Transient restlessness, insomnia.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Contraindicated in pregnancy. Lactation: Contraindicated or not recommended. Contraindications: The immediate recovery phase after myocardial infarction. Defects in bundle-branch conduction. Untreated narrow angle glaucoma. Acute alcohol, barbiturate and opiate intoxications. Patients who have received a MAO-inhibitor within two weeks. Excitable or overactive patients. Precautions: If previously the patient has been treated with tranquillizers with sedative effect these should be withdrawn gradually.

Interactions

Alcohol, barbiturates and other CNS depressants. MAO-inhibitors. Neuroleptics. Guanethidine. Adrenaline, Noradrenaline.

Advice to Patient

Take with or without meals.

FLUPENTHIXOL: Onset: Oral: Within 2-3 days. IM: 24-72 hrs. Duration: Oral: 8 hrs. IM: 2-4 wks. Absorption: Oral: Readily absorbed from the GI tract. IM: Slowly released from IM inj sites. Metabolism: Hepatic. Bioavailability: Oral: 40%. Half-life elimination: Oral: 35 hrs. IM: 3 wks. Time to peak, plasma: Oral: 4 hrs. IM: 4-7 days. Excretion: Urine. Faeces. MELITRACEN: Absorption: Well absorbed from the GI tract. Metabolism: Hepatic (undergoes extensive first-pass metabolism). Half-life elimination: Approx. 19 hrs. Time to peak, plasma: 4 hrs. Excretion: Urine and faeces.
