Deferiprone
Antidote · Oral iron chelator
Indications
Treatment of iron overload in patients with thalassaemia major for whom deferoxamine therapy is contraindicated or who present serious toxicity with deferoxamine therapy.
Dosage
Adults & Children: 25 mg/Kg of body wt three times daily. Max: 100 mg/Kg/day.
Adverse Effects
GI disturbances, joint pain. Rarely, agranulocytosis, neutropenia.
Risk Factor (Pregnancy/Lactation)
Contraindications: Neutropenia, agranulocytosis. Pregnancy & lactation. Precautions: Monitor neutrophil count, serum ferritin & Zn conc regularly. Discontinue if infection develops. Renal & hepatic impairment. Children.
Interactions
Aluminium containing antacids.
Pharmacokinetics
Absorption: Absorbed rapidly from the upper part of the GI tract. Metabolism: Hepatic. Half-life elimination: 1.9 hrs. Time to peak, serum: 1-2 hrs. Excretion: Urine (75%-90%).
