Cyclizine

Antiemetic & Gastroprokinetic · Piperazine derivative

Indications

Nausea, vomiting, travel sickness, vertigo.

Dosage

Adults: 50 mg up to three times daily. Children: Under 6 yr: Not recommended. 6-12 yr, 25 mg up to three times daily; 12-18 yr, 50 mg up to three times daily.

Adverse Effects

Tachycardia, urticaria, rash, headache, drowsiness, dry mouth, blurred vision, urinary retention, constipation, restlessness, nervousness, insomnia, auditory visual hallucinations. Rarely cholestatic jaundice, hepatitis, hypersensitivity reactions. Agranulocytosis, asthaenia, muscle spasms, bronchospasm, apnoea, hypertension, disorientation.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Contraindicated in pregnancy. Lactation: Caution advised or effect undetermined. Contraindications: Porphyria. Precautions: Glaucoma, severe heart failure, renal or hepatic impairment, GI obstruction, epilepsy, prostatic hypertrophy.

Interactions

Anticholinergics, alcohol, CNS disturbances, pethidine.

Advice to Patient

Avoid alcohol and other CNS depressants during therapy. If there is drowsiness or dizziness ask for help with ambulation.

Onset: Within 2 hrs. Duration: 4 hrs. Absorption: Absorbed from GI tract. Metabolism: Hepatic. Half-life elimination: 20 hrs. Time to peak, plasma: Approx. 2 hrs. Excretion: Urine (<1% of total oral dose).

Unlicensed Use

Children: Nausea and vomiting of known cause; nausea and vomiting associated with vestibular disorders and palliative care: 1 mth-6 yr, Orally 0.5-1 mg/kg up to 3 times daily, max single dose 25 mg.
