Indications

Gram-positive infections (including resistant staphylococci).

Dosage

ORAL: Adults: 500 mg six hourly. Children: Under 2 yr, quarter adult dose, over 2 yr, half adult dose. INJECTION: Adults: 250 mg IM four to six hourly. Children: Under 2 yr, quarter adult dose, over 2 yr, half adult dose.

Administration

For IM Injection: 250 mg VIAL/500 mg VIAL: Add 1.5 mL SWFI in 250 mg VIAL or 2 mL SWFI in 500 mg. For IV Injection: Add 5 mL SWFI in 250 mg VIAL or 10 mL SWFI in 500 mg VIAL, then administer by Direct IV Injection or by IV Intermittent Infusion after addition of appropriate infusion fluid. Stability: Store at controlled room temperature not exceeding 25°C. Reconstituted solution is stable for up to 24 hours at controlled room temperature and 48 hours under refrigeration.

Adverse Effects

Occasional rashes, GI upsets, anaphylaxis.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Risk cannot be ruled out. Lactation: Caution advised or effect undetermined. Contraindications: Hypersensitivity to penicillins. Precautions: Renal impairment. Newborns, hypersensitivity to penicillins.

Advice to Patient

Take 1 hour before or 2 hours after meals. Take with a full glass of water, do not take with fruit juice or soda. Use a calibrated measuring device to ensure accurate doses. Contact prescriber if signs of allergic reaction occur. Complete full course of therapy even if you feel better. Refrigerate oral solution container and discard unused portion after 14 days.

Duration: 6 hrs. Absorption: Incompletely absorbed from the GI tract. Absorption is reduced by the presence of food in the stomach. Metabolism: Undergoes limited metabolism. Bioavailability: Oral: 50%. Half-life elimination: 0.5-1 hr. Prolonged in neonates. Time to peak, plasma: Fasting patients: 1-2 hrs. Excretion: Urine (35%). Faeces (10%).
