Indications

INJECTION: CVA in acute & subacute phase. Recent head trauma & their sequela. Post-traumatic & post-operative disturbance of consciousness & motor paralysis after apoplexy. To improve brain metabolism in cerebrovascular disease. ORAL: Treatment of cognitive, sensitive, motor and neuropsychological disorders of degenerative or vasculocerebral aetiology.

Dosage

INJECTION: Adults: Usually 1000 mg-2000 mg daily IM or slow IV or IV infusion (infusion rate 40-60 drops/min), for four consecutive wk. Can be administered orally. Children: Not recommended. ORAL: Adults: 600-2000 mg daily. Children: Above 12 yr, 100 mg two to three times daily.

Administration

Administered by IM or IV (3-5 min) and IV drip (infusion rate of 40-60 drops/min). Direct IV administration should be made very slowly to prevent episodes of hypotension. Compatibility: Hypertonic glucose solution and all IV isotonic solns.

Adverse Effects

Inj: Hypotension and parasympathetic effects. Drop: GI disorders.

Risk Factor (Pregnancy/Lactation)

Contraindications: Parasympathetic hypertonia. Precautions: Persistent intracranial haemorrhage.

Interactions

Levodopa.

Advice to Patient

Take with or without meals. To prevent stomach upset, take with meals. Contact prescriber if there is headache, slow or fast heartbeat, nausea or vomiting, low blood pressure, faintness, dizziness, diarrhoea. Avoid alcohol during therapy.

Absorption: Rapidly absorbed. Metabolism: Hepatic. Also in gut wall. Bioavailability: >90%. Half-life elimination: 24 hrs. Time to peak, plasma: Biphasic. 1st peak: 1 hr after dosing. 2nd peak: 24 hrs. Excretion: Faeces (<1%).
