Indications

Adjunct in the treatment of female infertility.

Dosage

Adults: Females: By SC inj: Used in conjunction with controlled ovarian stimulation therapy using gonadotrophins (FSH, HMG). Single-dose regimen: 3 mg given when serum oestradiol levels show appropriate stimulation response, usually stimulation day 7 (range days 5-9). If hCG is not administered within 4 days, continue cetrorelix at 0.25 mg/day until hCG is administered. Multiple-dose regimen: 0.25 mg morning or evening of stimulation day 5, or morning of stimulation day 6, continue until hCG is administered. Children: Not applicable.

Administration

Administer by SC inj. The needle should be inserted completely into the skin at a 45-degree angle. Stability: Store in outer carton. Once mixed, solution should be used immediately. 0.25 mg vial: Store under refrigeration at 2°C-8°C (36°F-46°F). 3 mg vial: Store at controlled room temperature at 25°C (77°F); excursions permitted to 15°C-30°C (59°F-86°F).

Adverse Effects

Nausea, headache, injection site reactions, rarely hypersensitivity reactions, ovarian hyperstimulation.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Contraindicated in pregnancy. Lactation: Contraindicated or not recommended. Contraindications: Severe renal impairment. Precautions: Should only be prescribed by fertility specialist. Hepatic and renal impairment. Repeated cycles, history of or active allergic conditions.

Advice to Patient

Administer at same time each day. Contact prescriber if there is shortness of breath, abdominal pain, vaginal bleeding, pain, itching or signs of infection at injection site. Contact prescriber if a dose is missed, do not take double dose. Keep all appointments as scheduled. Take mild analgesic if headache occurs.

Onset: 1-2 hrs. Duration: 3 mg: = 4 days. 0.25 mg: 24 hrs. Absorption: Rapidly absorbed after SC inj. Metabolism: Via peptidases. Bioavailability: 85%. Half-life elimination: 3 mg inj: 60 hrs. Less with lower doses. Time to peak, plasma: 3 mg inj: 1.5 hrs. 0.25 inj: 1 hr. Excretion: Urine (as unchanged drug). Faeces (small amount as metabolites).
