Calcium Folinate (Calcium leucovorin)
Antidote · Antidote to folic acid antagonist
Indications
As an antidote or “rescue” after treatment with (high-dose) methotrexate. In the treatment of megaloblastic anaemias, owing to folate deficiency.
Dosage
Adults and Children: Antidote: To be adjusted to individual needs, provided that it corresponds to approximately 10 to 20 per cent of the methotrexate dose. See literature. Megaloblastic anaemias: Treatment, initial course of 10 mg daily for 10 to 15 days, but 1 mg daily is probably adequate, subsequent therapy and dosage modified to the haematological response.

Metabolism: Hepatic and intestinal mucosa. Half-life elimination: 6.2 hrs. Time to peak: 5-formyl-THF (parent compound): 10 mins. 5-methyl-THF (active metabolite): 1.3 hrs.
