Antihistamine, Systemic · H1 receptor antagonist

Indications

Seasonal and perennial allergic rhino conjunctivitis, urticaria.

Dosage

Tab: Adults: 1 daily one hour before or two hours after food. Children: Not recommended. Syp: Children 6-11yrs: 10 mg (4mL) once daily. >12yrs: 20 mg once daily.

Adverse Effects

Headache, drowsiness.

Risk Factor (Pregnancy/Lactation)

Pregnancy: Risk cannot be ruled out. Lactation: Contraindicated or not recommended.

Interactions

P-gp inhibitors (e.g. erythromycin, ketoconazole, ritonavir, diltiazem, ciclosporin), rifampicin, grapefruit juice.

Onset of action: 1 hr. Duration of action: 26 hrs. Absorption: Rapidly absorbed from the GI tract. Bioavailability: Approx. 61%; decreased with food. Metabolism: Minimal. Half-life elimination: Approx. 14.5 hrs. Time to peak, plasma: 1.13 hrs. Excretion: Faeces (66.5%). Urine (28.3%).
