Azathioprine

Indications
Transplantations; certain auto-immune diseases.
Dosage
Adults: Transplantation: Initially 5 mg/kg, then 1-4 mg/kg daily in divided doses for 3 mth. Chronic active hepatitis: 1-1.5 mg/kg daily. Other indications: 2-2.5 mg/kg daily. Children: Same as adults.
Adverse Effects
Myelosuppression, hepatotoxicity, Gl distress, rashes.
Risk Factor (Pregnancy/Lactation)
Pregnancy: Risk cannot be ruled out. Contraindications: Biliary stasis; toxic hepatitis. Precautions: Monitor for toxic effects, especially blood counts. Infection. Impaired renal function. Undue sun exposure.
Interactions
Allopurinol, cytostatics, muscle relaxants.
Pharmacokinetics
Absorption: Well-absorbed. Metabolism: Metabolized primarily to mercaptopurine. Both compounds are further metabolized in RBCs and liver. Half-life elimination: Approx. 5hrs. Time to peak, serum: 1-2 hrs. Excretion: Urine.
