Indications
Treatment of malaria.
Dosage
Acute attacks in non-immune subject, Adults: Single dose of 600 mg followed by 300 mg doses 6, 24 and 48 hr later. For partially immune subjects: Adults: Single dose of 600 mg is often sufficient. Children: Upto 1 yr, 50 mg; 1-4 yr, 50-100 mg; 5-8 yr, 150-200 mg; 9-12 yr, 200-300 mg. All once a wk.
Adverse Effects
Headache, Gl distress. Skin eruptions, depigmentation, hair loss, blurred vision, corneal opacities, retinal damage, blood disorders.
Risk Factor (Pregnancy/Lactation)
Pregnancy: Risk cannot be ruled out. Lactation: Contraindicated or not recommended. Precautions: Porphyria. Impaired renal or hepatic function. Psoriasis. Perform eye test before & during long-term therapy. Severe gastrointestinal, neurological or blood disorders.
Interactions
Antacids, kaolin, cimetidine, mefloquine, cyclosporin, rabies vaccine, anticoagulants, amiodarone, digoxin, neostigmine, pyridostigmine, moxifloxacin.

Absorption: Readily absorbed from the GI tract. Metabolism: Hepatic. Half-life elimination: 5.2 ± 1.7 mins. Excretion: Urine.
